Toxicity Screening & Selectivity Profiling of Cardiac Ion Channels

Over-dependence on the hERG assay as a preclinical cardiac risk indicator can terminate development of potentially useful compounds that non-selectively block hERG and other compensatory (e.g., voltage-gated Na+ and Ca2+) channels. Conversely, some compounds may pose a safety risk by prolonging repolarization via interactions with non-hERG cardiac channels. Automated patch clamp screening and concentration-response assays provide a rapid, comprehensive survey of cardiac channels for greater insight into potential cardiac risk and more effective decision-making in early preclinical development.
