  • Mustard-mediated calcium response of EZCellsTM compared to untransfected CHO-K1 Cells. Gamma-irradiated CHO cells that express the human TRPA1 channels (EZCellsTM, green circles) or untransfected CHO-K1 cells (black squares) were loaded with Fluo-4 NW (Invitrogen), then stimulated with the indicated concentrations of mustard oil. The resulting calcium responses were measured on a FlexStation fluorescent plate reader. Data represent the average ±standard deviation of 6 replicates.
  • Ruthenium red inhibition of mustard-induced calcium response. CHO cells that stably express the human TRPA1 channels (blue triangles) and similar cells that were irradiated as EZCellsTM (green circles) or untransfected CHO-K1 cells (black squares) were loaded with Fluo-4 NW (Invitrogen), then treated with the indicated concentrations of ruthenium red, then stimulated with 60 μM mustard oil. The resulting calcium response was measured on a FlexStation fluorescent plate reader. Data represent the average ± standard deviation of 6 replicates.

Quick Overview

Human TRPA1 ion channel-expressing, division-arrested cells.

Background

The human TRPA1 gene encodes the pore-forming subunit of TRPA1, a non-selective cation channel expressed in sensory neurons and hair cells. TRPA1 channels are therapeutic targets in treatment of pain.
