  • GABA-mediated membrane depolarization of EZCellsTM compared to non-irradiated cells. CHO cells that stably express the Human GABAA (α1β2γ2) Channels (blue triangles) and similar cells that were irradiated as EZCellsTM (green circles) or untransfected CHO-K1 cells (black square) were loaded with membrane potential dye (Molecular Devices), then stimulated with the indicated concentrations of GABA. The resulting membrane depolarization was measured on a FlexStation fluorescent plate reader. Data represent the average +/- standard deviation of 6 replicates.
  • Picrotixin inhibition of GABA-induced membrane depolarization. CHO cells that stably express the Human GABAA (α1β2γ2) Channels (blue triangles) and similar cells that were irradiated as EZCellsTM (green circles) or untransfected CHO-K1 cells (black square) were loaded with membrane potential dye (Molecular Devices), then treated with the indicated concentrations of picrotoxin and next stimulated with 10 uM GABA. The resulting membrane depolarization was measured on a FlexStation fluorescent plate reader. Data represent the average +/- standard deviation of 6 replicates.

Quick Overview

Human GABAA (α1β2γ2) ion channel-expressing, division-arrested cells.

Background

The human GABRA1, GABRB2, and GABRG2 genes encode the Cl--selective, ionotropic GABAA receptor-channel. GABAA receptors expressed in neurons are therapeutic targets in treatment of neurological disorders.
