  • Fluorescence measured upon channel activation with high K+ in the presence or absence of the Cav3.2 antagonist pimozide. Calcium 4 dye was loaded in 0 Ca2+/0.5 mM K+ HB-PS. Stimulation buffer included vehicle or pimozide and brought final Ca2+ and K+ concentrations to 6.75 mM and 84.6 mM, respectively. A: Average fluorescence signal for each test condition.  Pimozide at 6.3 μM and 100 μM blocked 51% and 94% of Cav3.2 signal, respectively (measured at maximum fluorescence signal). Recording instrument: FLIPR.

Background

The human CACNA1H gene encodes the pore-forming subunit of Cav3.2, a voltage-gated T-type calcium channel. Coexpression with the β2 and α2δ1 auxiliary subunits recapitulates function in heterologous expression (HEK cells). Inclusion of the inwardly rectifying potassium channel allows the membrane potential to be altered with changes in extracellular K+.
