  • Pharmacological characteristics of hNav1.1 recorded by IonWorks Quattro®: pre-conditioning depolarization enhances sensitivity (use-dependent block) to lidocaine and carbamazepine. Aa-Ad: Block of hNav1.1 channels by lidocaine and carbamazepine measured using the stimulus voltage pattern shown in Aa (holding potential, -80 mV). The pulse pattern was repeated twice: before and after five minutes of test article (TA) exposure. Peak current amplitudes were measured at the first test pulse (TP1, tonic block, panel Ab), the 11th test pulse (TP11, frequency-dependent block, panel Ac), and the 12th test pulse (TP12, inactivated state block, panel Ad) .
  • Lidocaine concentration-response relationship. IC50 values for tonic, frequency dependent, and inactivated state block, respectively, were 703 µM, 49 µM (10 Hz stimulation frequency), and 4 µM. Data are presented as mean ± SD (n = 4-8). Recording instrument: IonWorks Quattro®.
  • Carbamazepine concentration-response relationship. IC50 values for tonic, frequency-dependent, and inactivated state block were >300 µM, 102 µM (10 Hz stimulation frequency), and 10 µM. Data are presented as mean ± SD (n = 3-5). Recording instrument: IonWorks Quattro®.

Background

The human SCN1A gene encodes the pore-forming subunit of Nav1.1, a voltage-gated sodium channel expressed in CNS neurons and dorsal root ganglia. Mutations in SCN1A cause several types of epilepsies (generalized epilepsy with febrile seizures, severe myoclonic epilepsy in infancy) and some forms of familial hemiplegic migraine. Nav1.1 channels are therapeutic targets in seizure, stroke, and pain.
