  • Orphanin FQ-mediated inhibitation of intracellular cAMP accumulation of EZCells™ compared to non-irradiated cells. CHO-K1 cells that stably express the ORL1 opioid receptor (black triangles) and similar cells that were irradiated (green circles) were exposed to the indicated concentrations of Orphanin FQ for 20 min at 37°C along with 10 µM forskolin and 1 mM IBMX. Intracellular cAMP was assayed using the HitHunter Kit (GE Healthcare). Data points represent the average standard ± deviation of 4 determinations.

Background

Opioid receptors are widely distributed in brain, spinal cord and peripheral sensory and autonomic nerves. They are activated physiologically by the products of endogenous opioid peptide genes such as proenkephalin, podynorphin, pro-opiomelancortin and pronociceptin. Four opioid receptor subtypes have been identified including µ, δ, κ, and ORL1. All of these subtypes are coupled to cAMP modulation via Gi/o but each subtype has a distinct pharmacological profile.
