  • Radioligand binding to membranes from cells expressing hV2 receptor. A crude P1 membrane fraction was made from cells expressing the hV2 receptor. Membranes were incubated with the indicated concentrations of [3H]-8AVP alone (red circles) or along with 8 µM unlabeled 8-AVP to measure non-specific binding. After incubation, membranes were filtered over GF/B filters and counted. Data represent the average of duplicate determinations.
  • CHO-K1 cells that stably express the hV2 receptor were loaded with Fluo4NW, then stimulated with the indicated concentrations of 8-AVP. The resulting increase in intracellular calcium was measured on a FlexStation fluorescent plate reader. Data points represent the average +/- standard deviation of triplicate determinations.

Background

The pituitary hormone vasopressin (AVP) is a cyclic nonapeptide that acts by binding to a family of vasopressin receptors that includes the V1a, V1b, and V2 receptors. V2 receptors are expressed in kidney where vasopressin exerts its antidiuretic action.
