  • Activation of TRPA1 by 60 µM mustard oil and block by ruthenium red, recorded by FLIPR® intracellular Ca2+ assay. Two replicate plates tested at varying ruthenium red concentrations (vertical axis) at final DMSO concentrations of either 0.18% or 1.0%.
  • Concentration-response relationship for ruthenium red inhibition. The IC50 values were 0.04 µM and 0.06 µM, respectively at 1% and 0.18% DMSO. Recording instrument: FLIPR®.
  • Reference Compounds (agonist): Somatostatin EC50: 13.5 nM

Quick Overview

Human TRPA1 ion channel-expressing, stable replicating cell line.

Background

The human TRPA1 gene encodes the pore-forming subunit of TRPA1, a non-selective cation channel expressed in sensory neurons and hair cells. TRPA1 channels are therapeutic targets in treatment of pain.
