  • U50488-mediated inhibition of intracellular cAMP response of EZCells™. The irradiated CHO-K1 cells that stably express the human Kappa opioid receptors were stimulated with 10 µM forskolin and the indicated concentration of agonist for 20 min at 37oC in the presence of 0.5 mM IBMX. Intracellular cAMP was assayed using the HitHunter cAMP Kit (GE Healthcare). Data points represent the average ± standard deviation of 6 determinations.

Background

Opioid receptors are widely distributed in brain, spinal cord and peripheral sensory and autonomic nerves. They are activated physiologically by the products of endogenous opioid peptide genes such as proenkephalin, podynorphin, pro-opiomelancortin and pronociceptin. Four opioid receptor subtypes have been identified including µ, δ, κ, and ORL1. All of these subtypes are coupled to cAMP modulation via Gi/o but each subtype has a distinct pharmacological profile.
